К моменту появления первых симптомов заболевания и соответственно возможности его диагностики происходит потеря 50–60 % дофаминергических нейронов чёрной субстанции и снижение уровня дофамина в стриатуме на 70–80 %. Современная фармакотерапия болезни Паркинсона всё ещё остаётся преимущественно симптоматической и направлена на компенсацию дефицита дофамина в ЦНС. В связи с этим актуально создание новых противопаркинсонических средств, способных как замедлять нейродегенерацию, так и стимулировать нейрогенез. Белки семейства нейротрофинов являются эндогенными нейропротекторами, а также активируют репаративные процессы в повреждённой нервной ткани. В настоящем обзоре суммированы данные литературы о связи нейротрофинов с патогенезом болезни Паркинсона и об их антипаркинсоническом потенциале.</p></abstract><trans-abstract xml:lang="en"><p>Parkinson's disease is the second most common neurodegenerative disease, associated with the progressive loss of dopaminergic neurons in the substantia nigra. By the time the ﬁrst symptoms of the disease appear and, accordingly, the possibility of its diagnosis arises, 50–60 % of dopaminergic neurons in the substantia nigra are lost, and the dopamine level in the striatum decreases by 70–80 %. Modern pharmacotherapy for Parkinson's disease is still predominantly symptomatic and aimed at compensating for the dopamine deﬁciency in the CNS. In this regard, the development of new antiparkinsonian drugs capable of both slowing down neurodegeneration and stimulating neurogenesis is relevant. Proteins of the neurotrophin family are endogenous neuroprotectors and also activate reparative processes in damaged nervous tissue.
