Когнитивные нарушения и тревожность являются распространёнными побочными эффектами при химиотерапии противоопухолевыми препаратами, особенно при использовании доксорубицина (ДОКС). В связи с этим, существует необходимость в разработке экспериментальных моделей для оценки вызываемых ДОКС дозозависимых нарушений когнитивных функций и эмоционального поведения.</p><p>Цель работы — изучение дозозависимого действия антрациклинового противоопухолевого препарата доксорубицина (ДОКС) на формирование и воспроизведение памятного следа, тревожность и моторно-двигательные реакции в экспериментах на здоровых половозрелых крысах.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Эксперименты выполнены на аутбредных крысах-самцах, которым вводили ДОКС (2,0 и 3,0 мг/кг, внутрибрюшинно) 1 раз/неделю в течение 4 недель. Для регистрации ориентировочно-исследовательского поведения, оценки моторно-двигательных реакций и тревожного поведения использовали тесты «открытое поле», «вращающийся стержень», «горизонтальная перекладина» и «приподнятый крестообразный лабиринт» (ПКЛ). Влияние ДОКС на процессы обучения и воспроизведения памятного следа изучали в тесте условного рефлекса пассивного избегания (УРПИ).</p></sec><sec><title>Результаты</title><p>Результаты. ДОКС в дозах 2 и 3 мг/кг не влиял на поведение крыс в тесте «открытое поле» и не нарушал координацию движений в тестах «вращающийся стержень» и «горизонтальная перекладина». В тесте ПКЛ ДОКС (3 мг/кг) усиливал тревожную реакцию. ДОКС в дозе 2 мг/кг, не вызывающей тревоги, не влиял на обучение крыс в тесте УРПИ. Однако через 24 ч и 7 суток ДОКС (2 мг/кг) снижал латентное временя захода в тёмный отсек установки по сравнению с контролем, что указывает на нарушение воспроизведения УРПИ.</p></sec><sec><title>Заключение</title><p>Заключение. Полученные данные подтверждают обоснованность использования ДОКС (2 мг/кг) для моделирования когнитивных нарушений и тревожности на этапе доклинического изучения биологически-активных веществ.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Background</title><p>Background. Cognitive impairment and anxiety are common side effects of anticancer chemotherapy, particularly with doxorubicin (DOX). Therefore, there is a need to develop experimental models to assess DOX-induced dose-dependent impairments in cognitive function and emotional behavior.</p><p>The aim — study of the dose-dependent effect of the anthracycline antitumor drug doxorubicin (DOX) on the formation and reproduction of a memory trace, anxiety, and motor responses in experiments on healthy sexually mature rats.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The experiments were performed on outbred male rats that were administered DOX (2.0 and 3.0 mg/kg, intraperitoneally) once a week for 4 weeks. To record exploratory behavior, assess motor reactions and anxiety behavior, the following tests were used: “open field”, “rotary rod”, “horizontal bar” and “elevated plus maze” (EPM). The influence of DOX on the processes of learning and reproduction of a memory trace was studied in the test of the conditioned reflex of passive avoidance (CPAR).</p></sec><sec><title>Results</title><p>Results. DOX at doses of 2 and 3 mg/kg did not affect rat behavior in the open field test and did not impair motor coordination in the rotarod and horizontal crossbar tests. In the EPM test, DOX (3 mg/kg) increased anxiety response. DOX at a dose of 2 mg/kg, which does not induce anxiety, did not affect rat learning in the CPAR test. However, after 24 hours and 7 days, DOX (2 mg/kg) reduced the latency time of entry into the dark compartment of the setup compared to the control, which indicates a disruption in the reproduction of the CPAR.</p></sec><sec><title>Conclusion</title><p>Conclusion.
