Болезнь Паркинсона (БП) — нейродегенеративное заболевание, характеризующееся двигательными нарушениями. Используемая в настоящее время противопаркинсоническая фармакотерапия, несмотря на симптоматическое улучшение пациентов, сопряжена с выраженными побочными эффектами. Комбинация субстанций ладастена с фабомотизолом может обладать потенциальным нейропротекторным эффектом в условиях нейродегенерации, наблюдаемой при БП, благодаря своему мультитаргетному механизму действия.</p></sec><sec><title>Цель работы</title><p>Цель работы. Изучение потенциальной антипаркинсонической активности комбинации субстанций ладастена с фабомотизолом на модели</p><p>паракват-индуцированного паркинсонического синдрома (ПС).</p></sec><sec><title>Методы</title><p>Методы. ПС моделировали внутрибрюшинным введением мышам линии C57Bl/6 параквата (10 мг/кг) 1 раз каждые 3–4 дня на протяжении 21 дня. Животные были разделены на 4 группы: 1) пассивный контроль, 2) активный контроль, 3) комбинация ладастен + фабомотизол, 4) леводопа. Животные были протестированы в батарее поведенческих тестов на каждой неделе эксперимента.</p></sec><sec><title>Результаты</title><p>Результаты. Комбинация субстанций ладастена с фабомотизолом успешно устраняла все проявления ПС в батарее поведенческих тестов, причём наиболее выраженная разница по сравнению с активным контролем в большинстве тестов была обнаружена на 3-й неделе эксперимента. Так, на фоне данной комбинации время удержания в тесте «вращающийся стержень» было выше в 2,38 (p &lt; 0,0001) и в 1,5 раза (p &lt; 0,01) при постоянной и нарастающей скорости, время поворота и спуска в тесте «вертикальный стержень» сократилось в 1,4 (p &lt; 0,05) и 1,53 раза (p &lt; 0,001).</p></sec><sec><title>Заключение</title><p>Заключение. Комбинация ладастена с фабомотизолом снижала выраженность ПС, моделируемого внутрибрюшинным введением параквата, в батарее поведенческих тестов, при этом сопоставимо с эффектами леводопы.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Relevance</title><p>Relevance. Parkinson's disease (PD) is a neurodegenerative disease characterized by motor impairments. The currently used antiparkinsonian pharmacotherapy, despite the symptomatic improvement of patients, is associated with severe side effects. The combination of ladasten with fabomotizole may have a potential neuroprotective effect in the neurodegeneration conditions observed in PD, due to its multitargeting action.</p></sec><sec><title>Objective</title><p>Objective. The study explores the potential antiparkinsonian activity of a combination of ladasten and fabomotizole in paraquat-induced model of parkinsonian syndrome (PS).</p></sec><sec><title>Methods</title><p>Methods. PS was modeled by intraperitoneal administration of paraquat (10 mg/kg) 1 time every 3–4 days for 21 days. The C57Bl/6 mice were divided into 4 groups: 1) passive control, 2) active control, 3) ladasten + fabomotizole combination, 4) levodopa. The animals were tested in the battery of behavioral tests each week of the experiment.</p></sec><sec><title>Results</title><p>Results. The combination of ladasten with fabomotizole successfully treated all manifestations of PS in the battery of behavioral tests, and the greatest difference compared with the active control was found in most tests on the 3rd week of the experiment. Thus, the combination increased latency to fall in the «rotarod test» by 2,38 (p &lt; 0.0001) and 1.5 times (p &lt; 0.01) at fixed and accelerating speed, decreased the turning and descent time in the «pole test» by 1.4 (p &lt; 0.05) and 1.53 times (p &lt; 0.001).</p></sec><sec><title>Conclusion</title><p>Conclusion.
