Фактор роста нервов (nerve growth factor, NGF), обладающий нейропротекторными и нейрорегенеративными свойствами, перспективен для разработки на его основе препаратов для лечения постинсультного состояния. ФГБНУ «ФИЦ оригинальных и перспективных биомедицинских и фармацевтических технологий» был сконструирован и синтезирован димерный дипептидный миметик NGF, получивший лабораторный шифр ГК-2, который в условиях экспериментальной церебральной ишемии выраженно снижал объём инфаркта мозга и стимулировал нейро- и синаптогенез.</p></sec><sec><title>Цель</title><p>Цель. Целью настоящего исследования стало изучение действия ГК-2 в отношении экспериментальной постинсультной деменции и депрессии.</p></sec><sec><title>Методы</title><p>Методы. Ишемический инсульт моделировали с помощью окклюзии средней мозговой артерии (ОСМА) на крысах Wistar. ГК-2 вводили в дозе 0,5 мг/кг, внутрибрюшинно, в течение 21 дня после ОСМА. Через 30–40 дней после ОСМА кратковременную и долговременную память животных оценивали в тесте распознавания нового объекта, депрессивноподобное состояние – в тесте вынужденного плавания и тесте предпочтения раствора сахара.</p></sec><sec><title>Результаты</title><p>Результаты. У животных группы «ОСМА» по сравнению с ложнооперированными животными была статистически значимо ухудшена (на 80 %) кратковременная и долговременная память, а также наблюдались поведенческие признаки депрессивноподобного состояния (увеличение времени иммобильности на 50 % и снижение предпочтения раствора сахара на 30 %). Дипептид ГК-2 полностью предотвращал развитие всех этих нарушений.</p></sec><sec><title>Заключение</title><p>Заключение. Таким образом, дипептидный миметик NGF ГК-2 противодействует развитию когнитивных и психоэмоциональных нарушений в условиях экспериментального инсульта.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Relevance</title><p>Relevance. The nerve growth factor (NGF), possessing neuroprotective and neuroregenerative properties, holds promise for the development of medications for the treatment of post-stroke conditions. At the Federal Research Center for Innovator and Emerging Biomedical and Pharmaceutical Technologies, a dimeric dipeptide mimic of NGF with the laboratory code GK-2 was designed and synthesized. Under conditions of experimental cerebral ischemia, it significantly reduced the volume of brain infarction and stimulated neuro- and synaptogenesis.</p></sec><sec><title>Objective</title><p>Objective. The aim of this study was to investigate the effects of GK-2 on experimental post-stroke dementia and depression.</p></sec><sec><title>Methods</title><p>Methods. Ischemic stroke was induced by occlusion of the middle cerebral artery (MCAO) in Wistar rats. GK-2 was administered intraperitoneally at a dose of 0.5 mg/kg for 21 days post MCAO. Short-term and long-term memory of the animals were assessed 30–40 days post MCAO using the novel object recognition test. Depressive-like state was evaluated through the forced swimming test and sucrose preference test.</p></sec><sec><title>Results</title><p>Results. In animals subjected to MCAO, both short-term and long-term memory exhibited a statistically significant decline of 80 %, along with a depressive-like state characterized by a 50 % increase in total immobility time and a 30 % reduction in sucrose preference, when compared to the sham-lesioned group. Dipeptide GK-2 completely averted the onset of these impairments.</p></sec><sec><title>Conclusion</title><p>Conclusion.
